I would like to simulate the reflected light from a ColorChecker tile (or even build an entire card) and use different illuminants in the system using the standard options for the Source Color (under Color/Spectrum) using different XYZ Tristimulus Values for different white points. 

I have  reflectance spectrum using a spectrophotometer which uses D65 illuminant

  1. How you make a surface with a custom reflectance spectrum ?
  2. Do I need to make transform my spectrum for each different illuminant I want to try?

Is this even something that can … should be done in Zemax? maybe that should be the first question

You can define the “custom” reflectance spectrum of a surface using coatings. Open the COATING.DAT file that comes with OpticStudio and search for “TABLE”:

Your result will change with the source because of the source spectrum.

I think you don’t need to change the reflection vs wavelength for each source. If it was measured with a spectrometer, then a blank was taken before each measure, so the value is a ratio, which is “relatively” independent of the source.
