I am modeling a very large Fresnel lens based on your model sample Fresnel 1. I set the lens unit to meter and I would like to have 100 grooves per meter. Can I set the +depth/-Frequency parameter to -100?

Yes, you are correct. If you set your +Depth/-Frequency to -100, it will yield 100 grooves per radial lens unit. In this case the lens unit is meters, so it will correspond to 100 grooves per meter.
