Its difficult to say for certain without an example file, but I'd say both lens can affect the light rays.
OpticStudio uses a nesting rule, which can be found in the Help file, that states:
If a ray strikes more than one object at the exact same point in space; the last object listed in the NSC Editor determines the properties of the surface or volume at that point.
In other word, if the inner lens comes later in the NSC Editor, then both lenses affect the rays. Otherwise, only the largest lens affect the rays, I think.
